Karl Landsteiner initially studied chemistry and worked with eminent scientists like Emil Fischer and Arthur Hantzsch before focusing on pathology and immunology.

The Discovery of Blood Groups (1900–1901)

Before Landsteiner’s work, blood transfusions were highly dangerous because doctors did not understand why some transfusions caused severe—often fatal—reactions.

In 1900, while working at the Pathological Institute of the University of Vienna, Karl Landsteiner observed that when the red blood cells (RBCs) of one person were mixed with the serum of another, the RBCs sometimes clumped (agglutinated). Through systematic testing, he identified three distinct patterns of agglutination and classified human blood into three groups: A, B, and C (later renamed O). In 1902, two of his colleagues (von Decastello and Sturli) identified a fourth group, AB. Thus, the ABO Blood Group System was established — the foundation of safe blood transfusion therapy.

Significance of the Discovery

  • Landsteiner’s discovery explained why some blood transfusions were successful while others failed.
  • It paved the way for modern blood banking and transfusion medicine, enabling safe matching between donors and recipients.
  • The ABO system remains the most important blood group system in transfusion practice worldwide.

Later Contributions

In 1940, Landsteiner (with Alexander Wiener) discovered the Rh (Rhesus) blood group system, identifying the Rh factor (D antigen) — another critical advance in transfusion medicine and in understanding Hemolytic Disease of the Newborn (HDN). He also contributed to the understanding of polio, immunology, and infectious diseases.

Honors and Recognition

  • Nobel Prize in Physiology or Medicine (1930) – for the discovery of human blood groups (ABO system).
  • His work earned him the title “Father of Blood Group System” or “Father of Immunohematology.”
